Bal Fluid Collection

BAL fluid collection, or bronchoalveolar lavage fluid collection, is a sampling technique used to obtain material from the lower respiratory tract for biological analysis. During the procedure, a bronchoscope delivers sterile saline into a selected airway segment and then aspirates the fluid, recovering airway-lining liquid, immune cells, microorganisms, proteins, and other cellular components. Researchers analyze these samples to assess pulmonary inflammation, infection, immune responses, and tissue injury. In biology, BAL fluid collection supports studies of lung function and disease mechanisms, enables comparison of cellular or molecular profiles, and provides a minimally invasive approach for investigating respiratory disorders and experimental treatments.

We describe a research technique for fiberoptic bronchoscopy and bronchoalveolar lavage using low pressure suction. The technique is used to harvest immune cells from the lung bronchoalveolar surfaces. Local anesthetic and mild conscious sedation (midazolam) is used. Subjects tolerate the procedure well and experience minimal side effects.

Transgenic (Tg) mouse models of AD provide an excellent opportunity to investigate how and why Aβ or tau levels in CSF change as the disease progresses in human patients. Here, we demonstrate a refined cisterna magna puncture technique for serial CSF sampling from the mouse.

Use of Cerebral Open-Flow Microperfusion for the Longitudinal Collection of Interstitial Fluid in an Animal Model of Glioblastoma

In vivo, intercellular communications underpinning many central nervous system diseases, such as glioblastoma (GBM), are notoriously difficult to measure and characterize. Here, we describe procedures of cerebral open-flow microperfusion (cOFM) that can be employed to sample interstitial fluid components in a longitudinal animal model of GBM.

This protocol describes an improved technique for the abundant collection of cerebrospinal fluid (CSF) with no contamination from blood. With greater sample collection and purity, more analyses can be performed using CSF to further our understanding of diseases that affect the brain and spinal cord.
