Cell Invasion Assay

A cell invasion assay is an in vitro technique that measures the ability of cells to move through an extracellular matrix barrier, providing a model of tissue invasion and metastatic behavior. Typically, cells are placed in an upper chamber separated from a lower chamber by a porous membrane coated with matrix material; a chemoattractant in the lower chamber promotes directional movement, and invading cells are later stained and quantified. Researchers use this assay to study cancer cell dissemination, characterize molecular regulators of motility, and evaluate how drugs or genetic changes affect invasive capacity, supporting investigations of disease mechanisms and therapeutic responses.

Commonly used, highly accessible methods for examining cell migration and invasion in vitro are described. The first method is the cell wound closure assay that measures cell motility. The second method is the transwell migration and invasion assay that assesses the chemotactic and invasive capacity of cells.

Invasion Assay Using 3D Matrices

2023

The extracellular matrix (ECM) is a network of molecules that provide a structural framework for cells and tissues and helps facilitate intercellular communication. Three-dimensional cell culture techniques have been developed to more accurately model this extracellular environment for in vitro study. While many cell processes during migration through 3D matrices are similar to those required for movement across rigid 2D surfaces, including adherence, migration through ECM also requires cells...

In this protocol, we describe the dissection of placentae from the mouse on pregnancy d10.5, followed by isolation of trophoblast cells using a Percoll gradient. We then demonstrate use of the isolated cells in a matrigel invasion assay.
