Biotin Labeled Probes

Biotin-labeled probes are nucleic acid or molecular probes linked to biotin, a small vitamin-derived molecule, for detecting complementary targets in biological samples. They work through specific binding between the probe and its target, followed by high-affinity recognition of biotin by streptavidin or avidin, which can carry enzymes or fluorescent labels to produce a measurable signal. These probes support in situ hybridization, Southern and Northern blotting, microarray analysis, and other assays that localize or quantify DNA and RNA. Their strong, versatile detection system improves sensitivity while allowing researchers to study gene expression, chromosome organization, and molecular interactions.

In this video, we demonstrate the procedure to determine the helicase activity of a target protein to unwind the biotin-labeled dsRNA substrate. The activity of the enzyme was identified by analyzing the electrophoretic mobility shift, followed by a chemiluminescence assay using chemiluminescent enzyme-conjugated streptavidin.
