Microtitre Plate Format

Microtitre plate format is a standardized arrangement of multiple small wells in a rectangular plate, allowing biological samples and reactions to be handled in parallel while using minimal volumes. Each well functions as an individual reaction vessel, and automated pipetting, incubation, mixing, and optical measurement support consistent processing across the plate. In biology, this format enables high-throughput assays such as enzyme activity measurements, cell viability tests, immunoassays, and nucleic acid analysis, with absorbance, fluorescence, or luminescence commonly used to quantify results. Its scalability and compatibility with robotic liquid handling make it important for screening, experimental replication, and efficient analysis.

The assay describes a rapid means to measure early biofilm formation in bacteria and fungi. This method uses a microtiter plate as the substratum for microbial biofilm formation, and the biofilm is visualized using crystal violet strain. The assay provides either a qualitative or quantitative assay for early biofilm formation.

The uneven loss of medium from microtiter plates affects the reproducibility of uniform multicellular tumor spheroid formation. Improving culture conditions to reduce significant medium loss will improve the reproducibility of spheroid formation and the results of spheroid-based assays using the liquid-overlay technique.
