Newcastle Disease Virus

Newcastle Disease Virus (NDV) is an enveloped, negative-sense single-stranded RNA virus that primarily infects birds and can cause Newcastle disease, a major threat to poultry health and production. In susceptible hosts, its hemagglutinin-neuraminidase protein attaches to sialic acid receptors, while the fusion protein mediates entry; the viral RNA polymerase then copies the genome and directs synthesis of viral proteins in the cytoplasm. NDV biology supports surveillance, molecular diagnostics, and vaccine development, while also providing a model for studying paramyxovirus replication, host range, and viral evolution.

Here we provide a detailed procedure for production, purification, and quantification of high-titer recombinant Newcastle disease virus. This protocol consistently yields > 6 × 109 plaque-forming units/mL, providing virus quantities appropriate for in vivo animal studies. Additional quality control assays to ensure safety in vivo are described.

Optimization of Antigen Preparation for Hemagglutination Inhibition Assay of Newcastle Disease Virus Serology

This article presents a simplified method for preparing 4 hemagglutination units antigen for Newcastle disease virus serological testing. By accurately determining the hemagglutination titer of the antigen, combined with a more rigorous back-titration method, and a well-defined adjustment process, it enhances test efficiency, reduces false positives, and improves disease surveillance in poultry.

To investigate the blood-retinal barrier permeability and the inner limiting membrane integrity in animal models of retinal disease, we used several adeno-associated virus (AAV) variants as tools to label retinal neurons and glia. Virus mediated reporter gene expression is then used as an indicator of retinal barrier permeability.
