Ovarian Medulla

The ovarian medulla is the inner region of the ovary, distinguished from the follicle-rich cortex by its loose connective tissue and extensive network of blood vessels, lymphatic vessels, and nerves. Its stromal framework supports these conduits, enabling delivery of oxygen and nutrients to ovarian tissues and transport of hormones and metabolic products, while contributing to communication between the ovary and the rest of the reproductive system. In biology and histology, examining the ovarian medulla helps researchers identify normal ovarian architecture, understand tissue support during follicular and reproductive activity, and recognize structural changes associated with disease or aging.

Changes in ovarian blood flow during the preovulatory period are essential for ovulation. Doppler ultrasonography has been used in humans and large animals to assess ovarian hemodynamics and perfusion. A protocol is presented for applying Doppler ultrasonography to evaluate ovarian hemodynamics during the preovulatory and periovulatory phases in mice.

In this video, we describe a method for isolating and characterizing primary ovarian cancer cells from solid ovarian tumor tissue specimens. The epithelial cancer cells obtained using this protocol are suitable for understanding the processes that may lead to ovarian cancer initiation and development.

To study in vivo tumor growth and tumor microenvironment, we used a syngeneic and orthotopic mouse model of ovarian cancer in immunocompetent animals. We transduced a mouse tumor cell line (MOV1) with Katushka fluorescent protein (MOV1KAT) and here we show its orthotopic implantation in ovary and in vivo imaging.
