Size Selection Filtration

Size selection filtration is a separation technique that sorts biological particles, cells, or molecules according to their physical dimensions, making it useful for isolating specific sample components. A fluid passes through a membrane or porous material with defined pore sizes, allowing smaller particles to pass while retaining larger ones; pressure, gravity, or centrifugation can drive the flow. In biology, the method supports cell fractionation, removal of debris, concentration of microorganisms, and preparation of samples for downstream analysis. By providing a relatively simple and scalable form of separation, size selection filtration helps improve sample purity and enables more consistent experimental results.

Tangential flow ultrafiltration (TFU) is a recirculation method used for the weight-based separation of biosamples. TFU was adapted to size-select (1-20 nm diameter) and highly concentrate a large volume of polydisperse silver nanoparticles (4 L of 15.2 μg ml-1 down to 4 ml of 8,539.9 μg ml-1) with minimal aggregation.

This manuscript describes an automated gel size selection approach for purifying DNA fragments for next-generation sequencing. The Ranger Technology provides complete automation of the entire process of agarose gel loading, electrophoretic analysis, and recovery of targeted DNA fragments allowing for high-throughput and high quality next-generation sequencing libraries.
