Virgin Female Flies

Virgin female flies are adult females that have not mated, a defined reproductive state that is essential for controlling genetic and behavioral experiments in biology. Researchers obtain them by separating newly emerged females from males before mating, preserving an unmated baseline for subsequent crosses or treatments; after mating, sperm storage and reproductive changes can alter fertility and behavior. These flies support controlled studies of inheritance, courtship, mate choice, fecundity, and reproductive physiology, particularly in model organisms such as Drosophila. Accurate identification and handling improve experimental reproducibility by ensuring that observed outcomes reflect the planned mating history rather than unknown prior exposure.

Reverse genetic approaches have proven extremely useful for determining which genes underly resistance to vector pathogens in mosquitoes. This video protocol illustrates a method used by the James lab to inject dsRNA into female A. aegypti mosquitoes, which harbor the dengue virus. The technique for calibrating injection needles, manipulating the injection setup, and injecting dsRNA into the thorax is illustrated.

Drosophila Courtship Conditioning: A Method to Test Learning and Memory In Flies

This video describes a classical conditioning assay that tests learning and memory in Drosophila, called courtship conditioning. The assay is based on the reduction of male courtship after experiencing a rejection by a non-receptive premated female. The example protocol shows a setup for the procedure that can be used to assess short- and long-term memory.

Drosophila Egg-Laying Preference Assay: A Method to Test Decision-Making in Flies

2023

Drosophila females choose between substrates and select sites that are attractive for egg-laying. This video describes a behavioral assay that tests their decision-making, called the egg-laying preference assay. The featured clip demonstrates the procedure with a custom-built setup, which enables researchers not only to test the behavior of many females at the same time but is also compatible with video recording and optogenetic setups.
