Akt Pathway

The Akt pathway is a cell-signaling network that regulates survival, growth, metabolism, and proliferation, making it a central focus of cancer research. Activation typically begins when receptor tyrosine kinases stimulate phosphoinositide 3-kinase (PI3K) to generate PIP3 at the plasma membrane, recruiting Akt for phosphorylation by PDK1 and mTORC2. Activated Akt then modifies downstream proteins that promote cell-cycle progression, inhibit apoptosis, and alter cellular metabolism. Genetic changes affecting PI3K, Akt, or PTEN can produce persistent signaling, supporting tumor development and treatment resistance. Studying this pathway helps researchers identify biomarkers, understand cancer biology, and develop targeted therapies.
