Il-17-positive Th17 Cells

IL-17-positive Th17 cells are a subset of CD4+ T helper lymphocytes that produce interleukin-17 (IL-17), a cytokine involved in inflammation, tissue defense, and immune regulation. They develop when naïve T cells receive antigen receptor signals together with cytokines such as transforming growth factor-β and inflammatory mediators, activating transcriptional programs that promote Th17 differentiation and IL-17 secretion. In cancer research, these cells are studied for their context-dependent effects on tumor immunity: IL-17 can enhance inflammatory responses and immune-cell recruitment, yet may also support tumor-promoting inflammation, angiogenesis, or progression in some settings. Measuring Th17 abundance and function therefore helps clarify tumor microenvironment biology and potential immunotherapy targets.

This video demonstrates the procedure of inducing paralysis by transferring activated Th17 cells targeting aquaporin-4 into a mouse model. In the central nervous system, the activated Th17 cells interact with aquaporin-4-expressing astrocytes, triggering an immune response that leads to tail and limb paralysis in the mouse.

Isolation and Th17 Differentiation of Naïve CD4 T Lymphocytes

With effector functions distinct from other T cell subsets, Th17 cells have been centrally implicated in inflammatory autoimmunity. This in vitro Th17 differentiation protocol provides a means to determine whether naïve CD4+ T lymphocytes can differentiate into Th17 cells, and to further examine their role in autoimmunity and host response.
