Mass Cytometry Preparation

Mass cytometry preparation is the set of sample-processing steps used to ready cells or tissues for mass cytometry, enabling simultaneous measurement of many biomarkers at single-cell resolution in cancer research. The workflow typically involves obtaining a viable, representative suspension, removing debris, fixing and permeabilizing when needed, blocking nonspecific binding, and staining cells with antibodies labeled by distinct metal isotopes; during analysis, an inductively coupled plasma atomizes the cells and time-of-flight mass spectrometry measures the isotope signals. Careful preparation preserves cell identity and antigen accessibility, reduces background, and supports high-dimensional profiling of tumor, immune, and stromal populations, helping researchers characterize tumor microenvironments and treatment responses.
