Murine Tumor Model

A murine tumor model is an experimental system that uses mice to study tumor development, progression, treatment response, and interactions with the immune system. Researchers generate or transplant tumors in mice, then monitor changes in tumor growth, tissue pathology, molecular signaling, and host responses under controlled conditions. These models include syngeneic tumors in immunocompetent mice, xenografts using human cancer cells, and genetically engineered strains that develop tumors through defined mutations. In cancer research, murine tumor models help evaluate anticancer drugs, immunotherapies, and combination treatments while revealing mechanisms of tumor biology, treatment resistance, and potential toxicity before clinical studies.

This protocol describes a method for the detailed evaluation of leukocyte subsets within the tumor microenvironment in a mouse tumor model. Chemerin-expressing B16 melanoma cells were implanted subcutaneously into syngeneic mice. Cells from the tumor microenvironment were then stained and analyzed by flow cytometry, allowing for detailed leukocyte subset analyses.
