Tumor Stemness

Tumor stemness is the set of self-renewal, differentiation, and cellular plasticity properties that enable some cancer cells to sustain tumor growth and adapt to changing conditions. These cells can divide asymmetrically, producing one stem-like daughter cell and one more differentiated cell, while signaling networks such as Wnt, Notch, and Hedgehog help maintain their state. In cancer research, studying tumor stemness helps explain intratumoral heterogeneity, treatment resistance, metastasis, and disease recurrence. Characterizing these properties may support the development of therapies that target tumor-initiating cells alongside the broader tumor population and improve long-term treatment outcomes.

The generation and characterization of tumor specific T cells using humanized mice is described here. Human thymic tissue and genetically modified human hematopoietic stem cells are transplanted into immunocompromised mice. This results in the reconstitution of an engineered human immune system allowing for in vivo examination of anti-tumor immune responses.
