Rat-tail Collagen I

Rat-tail Collagen I is a purified extracellular matrix protein derived from rat tail tendons and widely used as a biologically relevant scaffold in cancer research. Its triple-helical molecules assemble into fibrils under neutral pH and physiological temperature, forming hydrated gels whose concentration and stiffness can be adjusted to model tissue-like environments. These collagen matrices support tumor-cell adhesion, migration, invasion, and interactions with stromal cells, enabling researchers to study how extracellular matrix structure influences cancer progression. Rat-tail Collagen I is also used in three-dimensional culture systems, organoid models, and testing of anti-invasive therapies, helping connect cellular behavior with tumor microenvironment mechanics.

Blood samples are useful for assessing biomarkers of physiological states or disease in vivo. Here we describe the methodology to sample blood from the lateral tail vein in the rat. This method provides rapid samples with minimal pain and invasiveness.

This article describes the experimental procedures used to prepare rat tail tendons for biomechanical and mechanobiological studies. Several features of the main steps in preparation are demonstrated, beginning with extraction, cross-sectional area measurement, rinsing and loading into the bioreactor chamber.

Here, we describe a simple tail vein blood sampling method in non-anesthetized rats using a vacuum extraction tube system. This method reduces the risk of direct exposure to blood and simplifies taking multiple samples from a single venipuncture.

Organotypic Collagen I Assay: A Malleable Platform to Assess Cell Behaviour in a 3-Dimensional Context

A method is described for the preparation of a 3-dimensional matrix consisting of collagen type I and primary human fibroblasts. This organotypic gel serves as a useful substrate to assess invasive cell migration because it mimics basic features of tissue stroma and is amenable to many forms of microscopy.
