Synergism Detection

Synergism detection is the process of determining whether two or more interventions produce a combined effect greater than expected from their individual effects. In cancer research, investigators test drug combinations across concentrations and compare the measured response with a reference model, such as Bliss independence, Loewe additivity, or a combination index, while accounting for dose and experimental variability. These analyses help distinguish synergy from simple additivity or antagonism in cell viability, apoptosis, or other treatment responses. Reliable detection supports rational combination therapy design, identifies potentially effective treatment pairs, and guides further validation in preclinical models.

Synergism is a useful mechanism where combining two or more drugs is more effective than each constituent used alone. Such combinations are also called supra-additive interactions. The drugs collectively enhance the final therapeutic effect by acting on different targets.
