Light Source

A light source is a device or material that produces electromagnetic radiation, providing the controlled illumination required for observation, measurement, and photochemical reactions. It generates light when energy excites atoms, molecules, or electronic states, which then release energy as photons with characteristic wavelengths; sources may provide broad-spectrum or narrowly selected radiation. In chemistry, light sources support spectroscopy by enabling absorption and emission measurements, while ultraviolet and visible radiation can initiate photochemical transformations. Choosing an appropriate wavelength, intensity, and stability improves analytical sensitivity, reaction control, and reproducibility, making light sources essential in chemical research and instrumentation.

Gas sampling from laboratory-scale flames with online analysis of all species by mass spectrometry is a powerful method to investigate the complex mixture of chemical compounds occurring during combustion processes. Coupled with tunable soft ionization via synchrotron-generated vacuum-ultraviolet radiation, this technique provides isomer-resolved information and potentially fragment-free mass spectra.

We present a protocol for the crystallization of proteins using the crystallization facility in the Research Complex at Harwell and subsequent in situ X-ray crystallographic data collection from crystals within the plates at Diamond's Versatile Macromolecular Crystallography in situ (VMXi) beamline.
