Organic Phase Extraction

Organic phase extraction is a separation technique that transfers compounds between immiscible liquid phases, typically an aqueous phase and an organic solvent, according to their relative solubilities. In a separatory funnel, mixing increases contact between the phases, while differences in polarity and partition coefficients drive a target compound into the phase where it is more soluble; the layers then separate by density. Acid-base adjustments can selectively convert compounds between charged and neutral forms, improving their distribution and enabling targeted isolation. This method is widely used to purify reaction products, prepare samples for analysis, and remove impurities in synthetic and analytical chemistry.

This video describes the extraction of circulating microRNA from plasma samples of colorectal cancer patients using organic extraction followed by glass-fiber column enrichment. miRNAs are stable in biological fluids such as serum and plasma, which renders them ideal circulating biomarkers for cancer diagnosis, prognosis, and treatment decision-making and monitoring. This method will help identify new circulating biomarkers capable of predicting clinical outcomes in patients.
