Photodiode Array Detection

Photodiode array detection is an optical measurement technique that records light intensity across multiple wavelengths simultaneously, making it valuable for identifying and quantifying chemical compounds. In a typical detector, a dispersed beam reaches an array of photodiodes, with each diode converting incident photons into an electrical signal; the resulting spectrum reflects wavelength-dependent absorbance or transmission. Coupled with liquid chromatography, photodiode array detection monitors analytes as they elute, supports compound identification through spectral profiles, and assesses peak purity. This broad spectral view improves analytical confidence and helps researchers characterize mixtures, reaction products, and pharmaceutical compounds efficiently.

Mismatches in human leukocyte antigen (HLA) sequences between organ donor and recipient pairs are the major cause of antibody-mediated rejection in organ transplantation. Here we present the use of custom antigen arrays that are based on individual donors' HLA sequences to probe anti-donor HLA alloantibodies in organ recipients.

This video demonstrates printed glycan array technology for analyzing circulating anti-glycan antibodies in mouse serum. The antibodies in the serum interact with specific glycans on the chip, which is confirmed by immunostaining followed by fluorescence scanning.

A Peptide Array-Based Detection of Anti-donor HLA Alloantibodies in Organ Recipients

2025

This video demonstrates an assay to detect anti-donor HLA alloantibodies in organ recipients. A peptide array is synthesized to represent potential epitopes for antibody-mediated organ rejection. The array is incubated with the recipient's serum, containing alloantibodies that bind to their target epitopes. Antibody binding is detected using chemiluminescence to identify donor-specific HLA epitopes that incite alloantibody reactivity.
