Beta-cyclodextrin Complexation

Beta-cyclodextrin complexation is a formulation strategy that uses beta-cyclodextrin, a cyclic oligosaccharide, to improve the pharmaceutical properties of poorly water-soluble compounds. Its hydrophobic inner cavity temporarily encloses suitable drug molecules through noncovalent host–guest interactions, while the hydrophilic outer surface promotes dispersion in aqueous environments. This process can increase apparent solubility, protect active ingredients from degradation, and support more consistent dissolution without chemically modifying the drug. In clinical pharmaceutical development, beta-cyclodextrin complexation aids the design of oral, injectable, and other dosage forms, potentially improving formulation performance and drug bioavailability.

The motion regimes of ibuprofen encapsulated in β-cyclodextrin nanosponges polymer network are investigated using pulsed-field-gradient spin-echo (PGSE) NMR technique. Synthesis, purification, drug loading, implementation of the NMR pulse sequence and data analysis to work out the mean square displacement of the drug at several observation times are described in detail.
