Pacemaker Implantation

Pacemaker implantation is a medical procedure that places a small electronic device in the body to regulate an abnormally slow or irregular heartbeat. The system uses a pulse generator connected to one or more leads, which sense cardiac activity and deliver electrical impulses when the heart’s intrinsic rhythm falls below a programmed threshold. Clinicians typically use pacemakers to treat conditions such as symptomatic bradycardia, conduction block, and some rhythm disorders, improving heart rate and reducing related symptoms. Implantation also provides a foundation for monitoring cardiac function and tailoring therapy through device programming and follow-up care.

This protocol describes how to produce functional sinus nodal tissue from murine pluripotent stem cells (PSC). T-Box3 (TBX3) overexpression plus cardiac Myosin-heavy-chain (Myh6) promoter antibiotic selection leads to highly pure pacemaker cell aggregates. These “Induced-sinoatrial-bodies” (“iSABs”) contain over 80% pacemaker cells, show highly increased beating rates and are able to pace myocardium ex vivo.

Bead implantation is a valuable tissue manipulation technique that can be used to interrogate developmental mechanisms by introducing alterations in local cellular environments.
