Adult Stem Cells

Adult stem cells are undifferentiated cells found in mature tissues that maintain tissue health by producing new cells throughout life. They self-renew through repeated cell division and can generate specialized progeny in response to signals from their local microenvironment, or stem cell niche; asymmetric division allows one daughter cell to retain stem cell properties while the other begins differentiation. In developmental biology, studying adult stem cells reveals how tissues balance renewal, maturation, and repair after injury. Research on hematopoietic, intestinal, and neural stem cells also supports disease modeling, tissue engineering, and regenerative medicine, while helping clarify how disrupted regulation contributes to aging and cancer.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Establishing culture systems for the expansion of adult neural stem cells (aNSCs) allows for the examination and application of aNSCs for therapy. The subcallosal zone (SCZ) has recently been recognized as a novel neuroblast-forming region in adult mice. Here, methods for the isolation, expansion, and differentiation of SCZ-aNSCs are described.

Neural stem cells were prepared from the hippocampus of adult non-hibernating yearling Arctic ground squirrels (AGS). These neural stem cells can be expanded through numerous passages, differentiated and maintained as a nearly 50:50 neuron to glial culture.
