Binary Transcription System

A binary transcription system is a genetic strategy that separates control of gene expression from the gene being expressed, allowing researchers to regulate specific cells or developmental stages with greater flexibility. It typically combines a driver component, such as a tissue-specific promoter that produces a transcriptional activator, with a responder component in which the desired gene is placed downstream of matching DNA-binding sites; expression occurs when the two components are brought together, often through genetic crossing. In developmental biology, this system enables precise labeling, activation, inhibition, or ablation of selected cell populations, supporting studies of lineage specification, tissue formation, and gene function.

Here, we present a method for generating tissue-specific binary transcription systems in Drosophila by replacing the first coding exon of genes with transcription drivers. The CRISPR/Cas9-based method places a transactivator sequence under the endogenous regulation of a replaced gene, and consequently facilitates transctivator expression exclusively in gene-specific spatiotemporal patterns.
