Rpe Cell Expansion

RPE cell expansion is the controlled propagation of retinal pigment epithelial cells in vitro to generate sufficient material for studying retinal development, tissue maintenance, and disease. The process typically involves isolating RPE cells, placing them on a suitable culture substrate, and providing defined nutrients and signaling conditions that support cell survival, proliferation, and retention of key epithelial characteristics. Expanded cultures can help researchers examine how RPE cells mature, interact with photoreceptors, and respond to developmental or disease-related signals. This approach also supports retinal organoid studies, cell-based modeling, and the evaluation of strategies for restoring damaged retinal tissue.

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
