Lr-white Embedding

LR White embedding is a tissue-preparation technique that encases biological specimens in a hard acrylic resin for microscopy, preserving structure for detailed analysis. After fixation and dehydration, LR White resin infiltrates the specimen and polymerizes to form a supportive block that can be trimmed and sectioned into thin or ultrathin slices. In developmental biology, this method enables light and electron microscopic examination of embryos, tissues, and cellular structures during growth and differentiation. Its relatively low-viscosity resin and compatibility with selected immunolabeling procedures can help preserve both tissue architecture and molecular targets, supporting studies of morphogenesis, cell organization, and developmental defects.

Here we present methodology for the production of a focal stroke in murine white matter by local injection of an irreversible endothelial nitric oxide synthase (eNOS) inhibitor (L-Nio). Presented are two stereotactic variations, retrograde neuronal tracing, and fresh tissue labeling and dissection that expand the potential applications of this technique.
