Cryoinjury Induced Heart Regeneration

Cryoinjury-induced heart regeneration is a biological repair process in which cardiac tissue recovers after controlled freezing damage, providing a model for studying how the heart replaces injured muscle. During the procedure, a cold probe creates localized myocardial injury, triggering inflammation, temporary scar formation, and activation of surviving cardiomyocytes that support new muscle formation. In developmental biology, this model helps researchers track the timing and cellular coordination of injury responses, cardiomyocyte proliferation, vascular repair, and scar resolution. Comparing regenerative and nonregenerative species or manipulating signaling pathways can reveal principles relevant to congenital heart disease, tissue engineering, and future regenerative therapies.

Zebrafish represents a valuable model to study the mechanisms of heart regeneration in vertebrates. Here, we present a protocol for induction of a heart infarct in adult zebrafish using cryoinjury. This method results in massive cell death within 20% of the ventricular wall, similar to that observed in mammalian infarcts.

Source: Dalkin, W., et al. The Fibular Nerve Injury Method: A Reliable Assay to Identify and Test Factors That Repair Neuromuscular Junctions. J. Vis. Exp. (2016)This video demonstrates a surgical procedure to induce fibular nerve injury in a mouse to study neuromuscular junction (NMJ) regeneration. The procedure involves exposing the fibular nerve, applying controlled pressure to induce injury, and ensuring the epineurium remains intact to support axonal regeneration. Over time, macrophages...
