Bovine Oocyte Culture

Bovine oocyte culture is the controlled maintenance and maturation of cow egg cells outside the ovary, providing a model for studying early development and reproductive biology. In vitro conditions supply nutrients, hormones, temperature, and gas concentrations that support oocyte metabolism and meiotic progression from the germinal-vesicle stage to metaphase II, when the cell can be fertilized. Researchers use cultured bovine oocytes to examine cytoplasmic and nuclear maturation, fertilization, embryo development, and factors affecting developmental competence. The approach also supports assisted reproduction, embryo production, livestock breeding research, and investigation of cellular mechanisms that influence early mammalian development.

Here, we present a protocol to significantly reduce the mitochondrial DNA copy numbers in a bovine oocyte (P < 0.0001). This method utilizes centrifugation and bisection to substantially reduce oocyte mitochondria and may allow for an increased chance of development in the reconstructed interspecies somatic cell nuclear transfer embryos.

In vitro culture of bovine ovarian cortex and the effect of nutritional Stair-step diet on ovarian microenvironment is presented. Ovarian cortex pieces were cultured for seven days and steroids, cytokines, and follicle stages were evaluated. The Stair-Step diet treatment had increased steroidogenesis resulting in follicle progression in culture.
