Mandible Decalcification

Mandible decalcification is the removal of mineralized calcium salts from the jawbone so its cellular and tissue structure can be examined with standard histological methods. Decalcifying agents such as ethylenediaminetetraacetic acid (EDTA) chelate calcium gradually, while acidic solutions dissolve mineral more rapidly; careful control helps preserve morphology and molecular components. In developmental biology, this preparation enables sectioning of embryonic and postnatal mandibles to study bone formation, cartilage development, tooth–jaw interactions, and tissue remodeling. The resulting sections support microscopic analysis of growth patterns and cellular organization, linking developmental processes to craniofacial structure and disease.

A technique to genetically manipulate epithelial cells within whole ex vivo cultured embryonic mouse submandibular glands (SMGs) using viral gene transfer is described. This method takes advantage of the innate ability of SMG epithelium and mesenchyme to spontaneously recombine after separation and infection of epithelial rudiments with adenoviral vectors.
