Tol2-free Zebrafish

Tol2-free zebrafish are genetically engineered zebrafish that retain a desired transgene or genome modification without an active Tol2 transposon system, providing cleaner models for developmental biology. During line generation, Tol2 transposase can transiently mobilize DNA into the zebrafish genome; breeding and genotyping then identify offspring that inherit the intended insertion while losing the transposase or other Tol2 components. These stable lines help researchers study gene function, cell lineage, and embryonic development while minimizing ongoing transposon-mediated rearrangement or remobilization, improving the interpretation and reproducibility of developmental phenotypes.

In vivo Electroporation of Morpholinos into the Adult Zebrafish Retina

A method to conditionally knockdown a target protein’s expression in the adult zebrafish retina is described, which involves intravitreally injecting antisense morpholinos and electroporating them into the retina. The resulting protein is knocked down for several days, which allows testing the protein’s role in the regenerating or intact retina.

miniCoopR Vector-based Transgenesis: A Technique to Screen Melanoma-inducing Genes in Zebrafish Tumor Model

This video describes a method to screen for the melanoma modifying genes using a transgenic zebrafish tumor model. When injected with a melanoma-inducing miniCoopR vector, the zebrafish develops melanomas and can therefore be used to study the genetic factors affecting tumor development in zebrafish.
