Apical Radial Glia

Apical radial glia are neural progenitor cells that line the ventricular surface of the developing central nervous system and generate much of the embryonic brain’s cellular diversity. They maintain apical-basal polarity, contact the ventricular lumen through apical junctions, and divide at the ventricular surface; asymmetric divisions can self-renew the progenitor pool while producing neurons or intermediate progenitors that migrate along radial processes. Their long fibers also provide scaffolds for neuronal migration, linking proliferation with cortical organization. Studying apical radial glia helps explain neurogenesis, cortical layering, and developmental disorders, while informing research on brain organoids, neural regeneration, and the evolutionary expansion of the mammalian cerebral cortex.

Here we provide a protocol for culturing rat cortical neurons in the presence of a glial feeder layer. The cultured neurons establish polarity and create synapses, and can be separated from the glia for use in various applications, such as electrophysiology, calcium imaging, cell survival assays, immunocytochemistry, and RNA/DNA/protein isolation.
