Dmpc Dmpe-dtpa

DMPC/DMPE-DTPA is a functional phospholipid membrane system that combines dimyristoylphosphatidylcholine with a DTPA-modified phosphatidylethanolamine to create a stable, metal-binding interface. In aqueous conditions, the amphiphilic lipids self-assemble into bilayers, while the exposed DTPA groups chelate metal ions through multiple donor sites without disrupting membrane organization. This combination allows researchers to control surface chemistry, incorporate metal ions, and tune interfacial properties in engineered membranes. DMPC/DMPE-DTPA systems support the design of liposomes, biosensing platforms, molecular probes, and other nanostructured materials requiring lipid assembly and selective metal coordination.

The goal of this experiment is to determine and control the size, shape and stability of self-assembled discotic amphiphiles in water. For aqueous based supramolecular polymers such level of control is very difficult. We apply a strategy using both repulsive and attractive interactions. The experimental techniques applied to characterize this system are broadly applicable.

High-throughput Crystallization of Membrane Proteins Using the Lipidic Bicelle Method

2012

Bicelles are lipid/amphiphile mixtures that maintain membrane proteins (MPs) within a lipid bilayer but have unique phase behavior that facilitates high-throughput screening by crystallization robots. This technique has successfully produced a number of high-resolution structures from both prokaryotic and eukaryotic sources. This video describes protocols for generating the lipidic bicelle mixture, incorporating MPs into the bicelle mixture, setting up crystallizations trials (manually as well...
