Droplet Sizing

Droplet sizing is the measurement and characterization of liquid droplet dimensions, typically summarized as a size distribution rather than a single diameter. In engineering, instruments determine this distribution by analyzing how droplets scatter light, alter an electrical signal, or are captured in images, with results influenced by sampling conditions, droplet shape, and concentration. These measurements help optimize atomizers, sprays, emulsions, fuel injection, pharmaceutical delivery, and agricultural applications because droplet size affects evaporation, transport, deposition, mixing, and process efficiency. Reliable sizing also supports equipment comparison, quality control, and prediction of performance across operating conditions.

We present protocols to be used in the measurement of spray droplet size from agricultural nozzles used in both aerial and ground based agrochemical applications. These methods presented were developed to provide consistent and repeatable droplet size data both inter- and intra-laboratory, when using laser diffraction systems.

The present protocol describes how to use oil red O to dye lipid droplets (LDs), calculate the size and number of LDs in a fatty acid-induced fatty hepatocyte model, and use BODIPY 493/503 to observe the process of small LDs fusing into large LDs by live cell imaging.

Phase-shift nanoemulsions (PSNE) can be vaporized using high intensity focused ultrasound to enhance localized heating and improve thermal ablation in tumors. In this report, the preparation of stable PSNE with a narrow size distribution is described. Furthermore, the impact of vaporized PSNE on ultrasound-mediated ablation is demonstrated in tissue-mimicking phantoms.

The assembly and positioning of the mitotic spindle depend on the combined forces generated by microtubule dynamics, motor proteins and cross-linkers. Here we present our recently developed methods in which the geometrical confinement of spherical emulsion droplets is used for the bottom-up reconstitution of basic mitotic spindles.
