Cardiac Specific Promoter

A cardiac specific promoter is a regulatory DNA sequence that drives gene expression preferentially in heart muscle cells, enabling researchers to control when and where a gene is active. It works when cardiac-enriched transcription factors bind promoter elements and recruit the transcriptional machinery, initiating RNA synthesis in suitable cellular and developmental contexts. In genetics, these promoters can direct reporter genes, therapeutic transgenes, or gene-editing components to cardiac tissue while limiting expression in other cell types. Their selectivity supports studies of heart development and function, disease modeling, and targeted genetic interventions, although promoter activity can vary with cell state, species, and experimental conditions.

Promoter expression analyses are crucial to improving the understanding of gene regulation and the spatiotemporal expression of target genes. Herein we present a protocol to identify, isolate, and clone a plant promoter. Further, we describe the characterization of the nodule-specific promoter in the common bean hairy roots.

This protocol describes how to produce functional sinus nodal tissue from murine pluripotent stem cells (PSC). T-Box3 (TBX3) overexpression plus cardiac Myosin-heavy-chain (Myh6) promoter antibiotic selection leads to highly pure pacemaker cell aggregates. These “Induced-sinoatrial-bodies” (“iSABs”) contain over 80% pacemaker cells, show highly increased beating rates and are able to pace myocardium ex vivo.

We describe here, the establishment and application of an Tg(Myh6-cre)1Jmk/J /Gt(ROSA)26Sortm38(CAG-GCaMP3)Hze/J (referred to as αMHC-Cre/Rosa26A-Flox-Stop-Flox-GCaMP3 below) mouse reporter line for cardiac reprogramming assessment. Neonatal cardiac fibroblasts (NCFs) isolated from the mouse strain are converted into induced cardiomyocytes (iCMs), allowing for convenient and efficient evaluation of reprogramming efficiency and functional maturation of iCMs via calcium (Ca2+) flux.
