Kidney Serum Microrna

Kidney serum microRNAs are small, noncoding RNA molecules detected in blood serum that reflect genetic regulation and cellular activity in the kidneys. They act by binding complementary sequences in messenger RNAs, usually reducing translation or promoting mRNA degradation, and can circulate in stable forms protected by extracellular vesicles or protein complexes. Measuring their abundance with techniques such as reverse transcription quantitative PCR or sequencing can reveal changes associated with kidney development, injury, inflammation, and disease. These molecules therefore support research into gene regulation and may provide minimally invasive biomarkers for diagnosis, prognosis, treatment response, and personalized approaches to renal disease.

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns—non-coding regions of a gene—or intergenic regions—stretches of DNA present between genes. Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA ends...

This article presents a protocol for detecting microRNA expression in the kidneys of an acute kidney injury mouse model using quantitative real-time reverse-transcription polymerase chain reaction. This protocol emphasizes an ischemic kidney injury mouse model and the careful extraction of microRNA samples.

microRNAs are involved in the pathogenesis of IgA nephropathy. We have developed a reliable method for detecting microRNA expression levels in the kidneys of an IgA nephropathy mouse model (HIGA mice). This new method will facilitate to check for miRNAs involvement in IgA nephropathy.
