Antibody Detection Assay

An antibody detection assay is a laboratory method that identifies and measures antibodies in a biological sample, helping determine whether an immune response has occurred. In a typical immunoassay, target antibodies bind a specific antigen immobilized on a surface; a labeled secondary antibody then recognizes the bound antibodies and produces a measurable color, fluorescence, or enzyme-based signal related to the amount present. In immunology and infection research, these assays support serological testing, pathogen exposure assessment, vaccine-response studies, and monitoring of disease progression. Their sensitivity and specificity depend on antigen selection, sample quality, assay controls, and interpretation of the resulting signal.

2025

Multiplex fluorescence immunoassays are becoming a reference for serology testing. Their application, however, is limited by the complexity and time-consuming bead-analyte coupling processes. This report presents a semi-automated, reproducible multiplex microsphere bead coupling protocol that is comparable to the standard method while considerably reducing time to results.

2025

The video demonstrates a hemagglutination inhibition assay for detecting serum antibodies against specific antigens. Confirmation of hemagglutination inhibition in the test well and hemagglutination in the control well is achieved through distinct visual patterns. This assay plays a crucial role in assessing immune response, vaccine effectiveness, and studying viral infections in virology and immunology.

In this video, we demonstrate an in vitro colorimetric assay to detect the presence of neutralizing antibodies against alkaline phosphatase-expressing recombinant adeno-associated virus serotype 6 (rAAV6) vectors in sheep serum samples. The assay utilizes the reaction between the AAV encoding a human placental alkaline phosphatase gene and its substrate to generate an insoluble purple product.
