Antibody Feeding Approach

The antibody feeding approach is a cell-based method for tracking the movement and fate of surface proteins in living cells, making it useful for studying receptor trafficking and host-pathogen interactions. In this assay, a labeled or unlabeled antibody binds an extracellular epitope at the cell surface, after which cells are incubated under defined conditions to permit internalization; subsequent acid washing, fixation, or secondary staining distinguishes surface-bound antibody from internalized signal. Researchers use the approach to measure endocytosis, recycling, and intracellular transport of immune receptors or pathogen-associated molecules, linking cellular trafficking to infection, signaling, and immune regulation.

An Antibody Feeding Approach to Study Glutamate Receptor Trafficking in Dissociated Primary Hippocampal Cultures

This article presents a method to study glutamate receptor (GluR) trafficking in dissociated primary hippocampal cultures. Using an antibody-feeding approach to label endogenous or overexpressed receptors in combination with pharmacological approaches, this method allows for the identification of molecular mechanisms regulating GluR surface expression by modulating internalization or recycling processes.

We describe a method to label protein on the surface of living neurons using a specific polyclonal antibody to extracellular epitopes. Protein bound by the antibody on the cell surface and subsequently internalized via endocytosis can be distinguished from protein remaining on, or trafficked to, the surface during the incubation.
