Eye Inflammation Imaging

Eye inflammation imaging refers to noninvasive and microscopy-based methods for visualizing inflammatory changes in ocular tissues, helping investigators assess disease without relying solely on tissue sampling. Techniques such as fundus photography, optical coherence tomography, fluorescein angiography, and fluorescence microscopy capture structural disruption, vascular leakage, immune-cell accumulation, or pathogen-associated signals through differences in light scattering, fluorescence, or dye movement. In immunology and infection research, these measurements help connect immune responses to retinal and uveal injury, monitor disease progression, evaluate treatment effects, and identify biomarkers that may support earlier diagnosis and more precise therapeutic studies.

We describe a non-invasive imaging method for distinguishing inflammatory stages. Systemic delivery of luminol reveals areas of acute inflammation dependent upon MPO activity in neutrophils. In contrast, injection of lucigenin allows for visualization of chronic inflammation dependent upon Phox activity in macrophages.

This protocol describes a detailed method for the long-term ex vivo culture and live imaging of a Drosophila imaginal disc. It demonstrates photoreceptor differentiation and ommatidial rotation within the 10 h period of live imaging of the eye disc. The protocol is simple and does not require expensive setup.

Overview In response to tissue injury and infection, mast cells initiate inflammation. Mast cells release chemicals that increase the permeability of adjacent blood capillaries and attract additional immune cells to the wound or site of infection. Neutrophils are phagocytic leukocytes that exit the bloodstream and engulf invading microbes. Blood clotting platelets seal the wound and fibers create a scaffold for wound healing. Macrophages engulf aging neutrophils to end the acute inflammatory...
