Fc Receptor Engagement

Fc receptor engagement is the binding of antibody constant (Fc) regions to Fc receptors on immune cells, a process that connects antigen recognition with cellular effector functions. When antibodies coat a microbe or infected cell, their Fc regions cluster receptors such as Fcγ receptors, triggering signaling pathways that can promote phagocytosis, antibody-dependent cellular cytotoxicity, cytokine release, or other responses; receptor activity depends on antibody isotype and receptor class. In immunology and infection research, this interaction helps explain how antibodies clear pathogens, shape inflammation, and influence disease outcomes. Measuring or manipulating Fc receptor engagement supports vaccine evaluation, therapeutic antibody development, and studies of immune evasion.

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.

Compared with traditional affinity chromatography using protein A agarose bead-packed columns, protein A membrane adsorbers can significantly speed laboratory-scale isolation of antibodies and other Fc fragment-expressing proteins. Appropriate analysis and quantification methods can further accelerate protein processing, allowing isolation/characterization to be completed in one workday, instead of 20+ work hours.
