Fluorescence Measurement Technique

Fluorescence measurement is a technique that detects and quantifies light emitted by fluorescent molecules after they absorb light at a shorter wavelength. A fluorophore enters an excited electronic state and releases part of that energy as longer-wavelength emission, which instruments measure as fluorescence intensity or spectrum. In immunology and infection research, fluorescent labels attached to antibodies, nucleic-acid probes, or cellular markers can reveal the presence, location, and relative abundance of pathogens, immune cells, or specific molecules. These measurements support immunoassays, microscopy, flow cytometry, and diagnostic analysis, enabling researchers to track host-pathogen interactions and evaluate immune responses.

Measurement of Extracellular Fluorescent Glucose Analog Depletion: A Surrogate Measurement Technique to Assess Glucose Uptake in Mouse Organs Ex Vivo

In this video, we demonstrate an ex vivo technique to measure extracellular glucose analog depletion in harvested mouse organs. It involves incubating the mouse tissue in a solution containing fluorescently-labeled deoxyglucose, which is taken up by the cells. The glucose analog uptake can be indirectly determined by measuring the fluorescence of the incubated medium.

Measuring Phagosome pH by Ratiometric Fluorescence Microscopy

Phagosomal pH influences phagosome maturation, oxidant production, phagosomal killing as well as antigen presentation. Here we describe a ratiometric method for measuring time-course and endpoint pH changes in individual phagosomes in living phagocytes using fluorescence microscopy.

In Situ Measurement of Vacuum Window Birefringence using 25Mg+ Fluorescence

Presented here is a method to measure the birefringence of vacuum windows by maximizing the fluorescence counts emitted by Doppler cooled 25Mg+ ions in an ion trap. The birefringence of vacuum windows will change the polarization states of the laser, which can be compensated by changing the azimuthal angles of external wave plates.
