Furared Am

Fura Red AM is a cell-permeable, ratiometric fluorescent indicator used to measure changes in intracellular calcium, a key signal in immune activation and host-pathogen responses. After entering cells, intracellular esterases cleave its acetoxymethyl groups, trapping the dye inside; calcium binding then alters its fluorescence, allowing calcium levels or fluxes to be estimated from signal changes, often through ratio-based imaging or flow cytometry. In immunology and infection research, Fura Red AM can help characterize calcium signaling in lymphocytes, phagocytes, and other responding cells, linking receptor stimulation or microbial exposure to functional outcomes such as activation, secretion, and cytotoxicity.

We present a protocol to determine the optimal extraction solvent to measure cortisol from koala fur. The solvents used in this protocol are methanol, ethanol and isopropanol. Determining an optimal extraction solvent will aid in reliably measuring fur to determine the impact of chronic stress on koalas.

Vagus nerve stimulation (VNS) has been shown to be effective as an adjunct treatment for treatment-resistant depression (TRD). VNS leads to antidepressive and antisuicidal effects and quality of life improvements. This protocol offers a step-by-step guide to managing and adjusting a vagus nerve stimulator for efficient treatment of TRD.

2014

Understanding the endogenous molecular changes in adult stem cells during aging requires isolating the cells of interest. The method described here presents a simple and robust approach to enrich for and isolate Drosophila intestinal stem cells and the enteroblast progenitor cells by FACS at any time point during aging.
