Skin Inflammation Assay

A skin inflammation assay is a laboratory method used to measure inflammatory responses in skin cells, tissues, or reconstructed skin models, helping researchers assess immune activity and tissue damage. These assays typically expose the model to an irritant, pathogen-associated stimulus, or inflammatory mediator and then quantify responses such as cytokine release, immune-cell activation, barrier disruption, or changes in gene expression. In immunology and infection research, the approach supports investigation of host–pathogen interactions, inflammatory skin disorders, and therapeutic responses. Results can clarify disease mechanisms and guide the evaluation of anti-inflammatory drugs, antimicrobial treatments, and skin-protective formulations.

Malassezia-Induced Skin Inflammation and Fungal Burden Analysis in a Murine Model

This video demonstrates Malassezia-induced skin infection in mice. Malassezia oil suspension is applied over the wounded ear, triggering inflammation and swelling of the ear tissue. Later, the ears are dissected, homogenized, and spread on an agar plate for growth. The number of colonies indicates the fungal burden resulting from skin inflammation.

Facilitating Drug Discovery: An Automated High-content Inflammation Assay in Zebrafish

Here we describe a novel high-content chemically induced inflammation assay aiming at the identification of immune-modulatory bioactives. We have successfully combined automated microscopy with custom developed software scripts enabling automated quantification of the inflammatory response as well as further data processing, analysis, mining, and storage.

We describe a non-invasive imaging method for distinguishing inflammatory stages. Systemic delivery of luminol reveals areas of acute inflammation dependent upon MPO activity in neutrophils. In contrast, injection of lucigenin allows for visualization of chronic inflammation dependent upon Phox activity in macrophages.
