Tumor-specific T Cells

Tumor-specific T cells are immune lymphocytes that recognize antigens produced by cancer cells and help distinguish malignant tissue from healthy cells. Their T-cell receptors bind tumor-derived peptides presented by major histocompatibility complex molecules, triggering activation, cytokine release, and targeted killing through cytotoxic molecules such as perforin and granzymes. In immunology and infection research, these cells are studied in tumor immunity, tumor-infiltrating lymphocytes, and adoptive cell therapies that expand or genetically enhance antitumor responses. Understanding their specificity and limitations, including tumor antigen loss and immune suppression, supports the development of more effective cancer immunotherapies.

This video demonstrates an in vitro assay to validate chimeric antigen receptor (CAR) T cell activity. B-cell lymphoma cells expressing CD19 antigen and cytoplasmic luciferase are co-cultured with CD19-specific CAR T cells. CAR binding to CD19 on a tumor induces the CAR T cells to release cytokines and toxins that cause tumor cell cytotoxicity. The CAR T cell activity is evaluated by quantifying the released cytokine and decreased luciferase-mediated bioluminescence in the tumor cells.

This video demonstrates a technique to study the in vivo localization of tumor-specific antibodies in a mouse tumor xenograft model. Upon subcutaneously injecting cancer cells into mice for tumor formation, fluorescently-labeled antibodies are injected that bind to tumor-specific receptors, enabling in vivo assessment of antibody localization via imaging.
