Air Pollution Exposure

Air pollution exposure is the inhalation or contact with harmful airborne substances, including particulate matter, gases, and biological pollutants, that can affect human health. In the respiratory tract, deposited particles and reactive chemicals can injure epithelial cells, trigger oxidative stress and inflammation, and allow some pollutants to enter the bloodstream, influencing organs beyond the lungs. In medicine, studying exposure helps clinicians and researchers assess risks for respiratory and cardiovascular disease, recognize vulnerable populations, and connect environmental conditions with symptoms and disease progression. Exposure assessment also supports prevention strategies, public health guidance, and research on interventions that reduce pollution-related harm.

Pulmonary epithelial cells can be isolated from the respiratory tract of mice and cultured at air-liquid interface as a model of differentiated respiratory epithelium. A protocol is described for isolating, culturing and exposing these cells to mainstream cigarette smoke, in order to study molecular responses to this environmental toxin.

Source: Villela, H. D. M., et al. Prospecting Microbial Strains for Bioremediation and Probiotics Development for Metaorganism Research and Preservation. J. Vis. Exp. (2019).This video demonstrates the isolation of pollutant-degrading marine bacteria through serial dilution, colony selection, and growth in EE2-containing medium. By using EE2 as the sole carbon source, the protocol identifies bacterial strains capable of metabolizing this environmental pollutant.
