Cancer Cell Lines

Cancer cell lines are laboratory-maintained populations of tumor-derived cells that provide reproducible models for studying cancer biology and evaluating medical treatments. Researchers grow these cells under controlled culture conditions, where their ability to proliferate continuously reflects tumor-associated genetic and cellular changes, while defined media and environmental conditions support survival and expansion. Cancer cell lines help investigate signaling pathways, drug responses, treatment resistance, and interactions between cancer-related processes. They support early-stage drug screening and hypothesis testing, although differences from patient tumors mean that findings require confirmation in primary cells, organoids, animal models, or clinical studies.

Pancreatic cancer stem cells (CSCs) can be expanded in vitro using the anchorage-independent sphere culture technique, which represents a powerful tool to study CSC biology and can serve as the first step to develop novel CSC-targeting therapies. Here the methodology for expanding, analyzing and targeting of pancreatic CSCs is provided.

Cancer stem cells (CSCs) have been identified in a number of malignancies. In this protocol we describe a flow cytometric method utilizing aldehyde dehydrogenase activity and CD44 and CD24 expression to isolate CSCs from human pancreatic adenocarcinoma xenografts. These viable cells can then be used in functional and analytical studies.
