Mouse Heart Surgery

Mouse heart surgery is a group of microsurgical procedures used to alter, repair, or assess the heart in a living mouse, providing controlled models of cardiovascular disease. Under anesthesia, researchers open the chest through a thoracotomy, expose the heart, and perform interventions such as coronary artery ligation, myocardial injection, or vascular manipulation before closing the incision and supporting recovery. These procedures model myocardial infarction, ischemic injury, cardiac remodeling, and regenerative therapies with precise control over timing and tissue damage. The resulting models help evaluate disease mechanisms, biomarkers, drug candidates, gene therapies, and emerging cardiac devices in preclinical medicine.

A step-by-step video protocol of apical resection is demonstrated in this study. Apical resection is a recently highlighted surgical approach in mammalian heart regeneration research. This study may promote the application of apical resection as a standard methodology in research into the mechanism underlying heart regeneration.

Developmental studies in the mouse are hampered by the inaccessibility of the embryo during gestation. To promote the long-term culture of the embryonic heart at late stages of gestation, we developed a protocol in which the excised heart is cultured in a semi-solid, dilute Matrigel.

The cuff technique shortens and facilitates the model of heterotopic cervical heart transplantation in mice by avoiding technically challenging suture anastomoses of small vessels. The technical details shown in this video paper should allow researches to establish this model in their laboratories.
