Nanobody Validation

Nanobody Validation is the systematic assessment of whether a nanobody, a small antibody-derived binding domain, recognizes its intended molecular target with sufficient selectivity, affinity, stability, and biological activity for a defined use. It combines biochemical and cell-based tests, such as antigen-binding assays, competition or specificity measurements, thermal-stability analysis, and functional experiments, to distinguish genuine target engagement from nonspecific binding or loss of activity. In medicine, rigorous validation supports the development of nanobodies as diagnostic reagents, imaging probes, and therapeutic candidates, while improving reproducibility and helping researchers select formats suitable for challenging targets, intracellular delivery, or repeated clinical testing.

Live-cell Imaging of Endocytic Transport using Functionalized Nanobodies in Cultured Cells

Endocytic and retrograde transport of proteins from the plasma membrane to the trans-Golgi network is essential to maintaining membrane homeostasis and regulating signaling. Here, we describe a method to image and quantify endocytic transport of transmembrane cargo proteins by live-cell microscopy using derivatized anti-GFP nanobodies in HeLa cells.
