Oral Intralipid Tolerance Test

The Oral Intralipid Tolerance Test is a controlled clinical or research procedure that evaluates how an individual responds to a standardized oral lipid load, providing information about gastrointestinal tolerance and postprandial fat handling. After Intralipid administration, investigators monitor symptoms and collect timed physiological measurements, commonly including blood lipid concentrations, to characterize the rise and clearance of circulating triglycerides. The resulting response profile can help assess lipid absorption and metabolism, compare metabolic function between groups, and support studies of obesity, diabetes, dyslipidemia, and related disorders. As a standardized challenge, the test also provides a practical framework for investigating altered fat processing in medicine.

The current article describes the generation and metabolic characterization of high-fat diet-fed mice as a model of diet-induced insulin resistance and obesity. It further features detailed protocols to perform the oral glucose tolerance test and the insulin tolerance test, monitoring whole-body alterations of glucose metabolism in vivo.

The following protocol describes a novel method for chronic oral drug administration using an orally dissolving strip (ODS) in lieu of the more commonly used oral gavage method. We demonstrate that preclinical, oral drug delivery using the ODS method represents a safe, convenient, and humane alternative to oral gavage.

(2018)This video demonstrates the viability assessment of a synthetic oral microbial community during simulated oral, gastric, and intestinal digestion. Bacterial survival is quantified after each phase, revealing a progressive decline due to digestive stress.

Begin with a culture of Alishewanella, a Gram-negative, salt-tolerant bacterium.Next, streak the culture onto agar plates containing increasing concentrations of sodium chloride to simulate varying levels of saline stress.Overlay each plate with a layer of unsolidified medium to prevent oxygen diffusion and maintain the culture in an oxygen-limited environment.Seal and incubate the plates at an optimum temperature to support bacterial growth.During incubation, the bacterium exhibits varying...
