Orthogonal Measurements

Orthogonal measurements are independent ways of assessing the same biological feature, using different instruments, assays, or physical principles to reduce reliance on a single result. Because each method has distinct sources of error and susceptibility to interference, agreement between measurements provides cross-validation, while discrepancies can reveal bias, sample effects, or limitations in an assay. In medicine, this approach supports biomarker validation, diagnostic testing, and treatment monitoring by strengthening confidence in clinically relevant findings. Combining complementary measurements can also clarify disease mechanisms and improve the reliability, reproducibility, and interpretation of biomedical research.

Optimized sampling protocols and the development of new wipe materials can be facilitated by standardized measurements of collection efficiency from wipe-sampling. Our approach for sampling trace explosives uses an automated device to control speed, force, and distance during wipe-sampling followed by extraction of collected explosives.

Resonance Fluorescence of an InGaAs Quantum Dot in a Planar Cavity Using Orthogonal Excitation and Detection

Resonant excitation of a single self-assembled quantum dot can be achieved using an excitation mode orthogonal to the fluorescence collection mode. We demonstrate a method using the waveguide and Fabry-Perot modes of a planar microcavity surrounding the quantum dots. The method allows complete freedom in the detection polarization.
