Rodent Surgical Protocol

A rodent surgical protocol is a standardized set of procedures for planning, performing, and monitoring surgery in laboratory rodents, supporting consistent and ethically responsible medical research. It typically combines anesthesia, aseptic preparation, tissue handling, hemostasis, wound closure, and perioperative monitoring to control physiological stress and promote recovery while maintaining reproducible experimental conditions. In medicine, these protocols guide procedures used to model disease, evaluate surgical techniques, study tissue responses, and test therapeutics or devices. Careful protocol design and postoperative assessment improve animal welfare, data quality, and the translation of findings from preclinical studies.

Genetic crosses of rodent malaria parasites are performed by feeding two genetically distinct parasites to mosquitoes. Recombinant progeny are cloned from mouse blood after allowing mosquitoes to bite infected mice. This video shows how to produce genetic crosses of Plasmodium yoelii and is applicable to other rodent malaria parasites.

Source: Kay Stewart, RVT, RLATG, CMAR; Valerie A. Schroeder, RVT, RLATG. University of Notre Dame, IN The Guide for the Care and Use of Laboratory Animals1 dictates that rodent survival surgery be performed aseptically. Aseptic technique utilizes specific practices that minimize the contamination of the surgical site, including patient preparation, surgeon preparation, sterilization of instruments and other supplies, and the use of a clean and controlled environment.
