Therapeutic Strategy Testing

Therapeutic strategy testing is the systematic evaluation of treatment approaches to determine whether they are effective, safe, and suitable for a specific medical condition. It typically compares one or more interventions with an appropriate control while measuring predefined outcomes such as disease response, adverse effects, and patient functioning. In medicine, this process can assess drugs, combinations, dosing plans, behavioral interventions, or other clinical strategies across preclinical and clinical research. Reliable testing guides evidence-based treatment decisions, identifies limitations and risks, and supports the development of more effective, targeted approaches to patient care.

Barnes Maze Testing Strategies with Small and Large Rodent Models

The dry-land Barnes maze is widely used to measure spatial navigation ability in response to mildly aversive stimuli. Over consecutive days, performance (e.g. latency to locate escape cage) of control subjects improves, indicative of normal learning and memory. Differences between rats and mice necessitate apparatus and methodology changes that are detailed here.

Set-shifting, a form of behavioral flexibility, requires an attentional shift from one stimulus dimension to another. We extended an established rodent set-shifting task1 by requiring attention to different stimuli according to context. The task was combined with specific lesions to identify neuron subtypes underlying a successful shift.

Orthotopic Xenografting of Human Luciferase-Tagged Malignant Peripheral Nerve Sheath Tumor Cells for in vivo Testing of Candidate Therapeutic Agents

A method for reliably grafting luciferase-tagged human malignant peripheral nerve sheath tumor cells into the sciatic nerve of immunodeficient mice is described. The use of bioluminescence imaging to demonstrate proper establishment of tumor grafts and criteria for random segregation of animals into study groups are also discussed.

Laboratory Scale Production and Purification of a Therapeutic Antibody

This protocol describes the production of a therapeutic antibody in a mammalian expression system. The methods described include preparation of vector DNA, stable transfection and serum-free adaptation of a human embryonic kidney 293 cell line, set up of large scale cultures and purification using affinity chromatography.
