Tumor Xenografts

Tumor xenografts are experimental models in which tumor cells or tissue from one species, commonly human cancer cells, are implanted into an animal to study cancer in a living system. Typically, researchers introduce tumor material into immunodeficient mice, allowing it to engraft and grow with reduced rejection by the host immune system. Cell-line xenografts provide reproducible models for investigating tumor growth and drug response, while patient-derived xenografts can preserve characteristics of an individual’s tumor. These models support preclinical evaluation of anticancer therapies, analysis of tumor biology, and investigation of treatment sensitivity, although differences between animals and patients can limit clinical translation.

In Vivo Fluorescence Imaging to Localize Antibodies in a Mouse Tumor Xenograft Model

2025

The video demonstrates a fluorescence imaging method for in vivo antibody localization in a mouse tumor xenograft model. It involves the injection of tumor cells in a mouse to generate solid xenografts beneath the skin. Subsequent injection of fluorescent antibodies confirms specific localization through enhanced fluorescence signals within the tumor xenograft.

DCE-MRI of Orthotopic Pancreatic Tumor Xenografts: A Method to Assess Microvasculature in a Target Tumor Tissue

2023

This video describes the technique of DCE-MRI for orthotropic pancreatic tumor xenografts in mice. DCE-MRI is a non-invasive method to analyze microvasculature in a target tissue, and useful to assess vascular response in a tumor following a novel therapy. This method will assist investigators to apply DCE-MRI for orthotopic gastrointestinal cancer mouse models.

This manuscript details a method used to generate prostate cancer patient derived xenografts (PDXs) from circulating tumor cells (CTCs). The generation of PDX models from CTCs provides an alternative experimental model to study prostate cancer; the most commonly diagnosed tumor and a frequent cause of death from cancer in men.

Tumor Enzymatic Digestion: A Procedure to Generate Tumor Cell Suspension from Patient-derived Xenograft Cancer Mouse Models

Mantle cell lymphoma (MCL) is a difficult to treat B cell disorder and it is equally difficult to establish a xenograft mouse model of primary MCL to study and develop therapeutics. Here, we describe the successful establishment of MCL xenografts in mice to help understand its underlying biology.
